STAT3-luc報(bào)告基因慢病毒是翊圣生物自主研發(fā)的用于檢測STAT3轉(zhuǎn)錄活性水平為目的的報(bào)告基因慢病毒顆粒。STAT3(signal transducers and activators of transduction-3)報(bào)告基因與多種惡性腫瘤的發(fā)生、發(fā)展和侵襲轉(zhuǎn)移關(guān)系密切。
STAT3報(bào)告基因慢病毒主要用于監(jiān)控細(xì)胞內(nèi)的STAT3轉(zhuǎn)錄活性、腫瘤****的研發(fā)以及基因過表達(dá)和RNAi的表型分析等。
pGMLV-STAT3-Lu是在pGMLV-Lu慢病毒載體的多克隆位點(diǎn)插入了多個(gè)STAT3結(jié)合位點(diǎn)，可以高靈敏度地檢測STAT3的激活水平。采用慢病毒作為報(bào)告基因的優(yōu)點(diǎn)在于它能夠感染多種難感染的細(xì)胞，比如原代細(xì)胞、干細(xì)胞、神經(jīng)元細(xì)胞等。而且慢病毒能夠?qū)⑼庠椿虿迦爰?xì)胞基因組內(nèi)，實(shí)現(xiàn)穩(wěn)定轉(zhuǎn)染。Lentivirus STAT3 Luciferase Reporter STAT3-Luc 報(bào)告基因慢病毒顆粒

使用說明
STAT3報(bào)告基因慢病毒顆粒采用慢病毒轉(zhuǎn)染方法轉(zhuǎn)染哺乳動(dòng)物細(xì)胞。用熒光素酶檢測試劑盒或雙熒光素酶檢測試劑盒進(jìn)行檢測。IL-6、白血病抑制因子（LIF）和血小板生長因子（PDGF）等可激活STAT3，可作為STAT3報(bào)告基因的陽性對照。
注意事項(xiàng)
1）病毒操作時(shí)*好使用生物**柜，如使用普通超凈工作臺(tái)操作病毒，請不要打開風(fēng)機(jī)。
2）病毒操作時(shí)請穿實(shí)驗(yàn)服，帶口罩和乳膠手套。
3）操作病毒時(shí)必須特別小心，不要產(chǎn)生氣霧或飛濺。如操作時(shí)超凈臺(tái)有病毒污染，立即用10%次氯酸鈉溶液搽拭干凈。接觸過病毒的槍頭、離心管和培養(yǎng)板等需用10%次氯酸鈉溶液浸泡1h以上后棄去。
4）用顯微鏡觀察細(xì)胞感染情況時(shí)應(yīng)遵從以下步驟：擰緊培養(yǎng)瓶或蓋緊培養(yǎng)板。用70%乙醇清理培養(yǎng)瓶外壁后到顯微鏡處觀察拍照。離開顯微鏡試驗(yàn)臺(tái)前，用70%乙醇清理實(shí)驗(yàn)臺(tái)。
5）病毒操作完成后，用肥皂清洗雙手。
